I would go the DI / IEM route. The less strain you put on your heart, the better.

I don't know about higher price indicating higher quality but I do know that a good set-up on any bass can make it feel like a million bucks.

At least .130 gauge for the B and a correctly cut nut will do wonders.

If it were me, I would be looking at a 300w combo, ideally a 1x15 or a 2x10. That way, it covers both practice and gigs. You will get far more value for your money secondhand. Alter Bridge December 2019 at London O2. Good band but just didn't do it for me live. Similar to Foo Fighters - songs played perfectly but nothing dangerous about them at all.

I think the cryogenic treatment is a gimmick and doesn't really improve the longevity that much. Dean Markley strings sound pretty broken in from the start so if you're after that sort of tone, GHS Boomers or black label Dunlop nickels might be a good alternative.
